HPLC-ELSD法测定L-抗坏血酸棕榈酸酯

摘要:建立高效液相色谱—蒸发光散射检测法检测L-抗坏血酸棕榈酸酯的方法。L-抗坏血酸棕榈酸酯样品用甲醇溶解,0.22滋m微滤膜过滤,仪器条件:用C18反相色谱柱进行分离,流动相:甲醇和水(含0.01%甲酸),流速:0.8mL/min,漂移管温度60℃,氮气压力:25psi,梯度洗脱。实验结果表明L-抗坏血酸棕榈酸酯峰面积的常用对数与其进样浓度的常用对数呈现良好线性关系(R2=0.9972),加标回收率在97.23%~102.05%范围内,RSD<5%。该方法不需对样品进行衍生化处理,能准确、快速定量L-抗坏血酸棕榈酸酯含量。

Determination of Ascorbyl Palmitate by HPLC-ELSD

Abstract: A high performance liquid chromatography with evaporative light-scatter detector(HPLC-ELSD)method forascorbyl palmitate was established. Ascorbyl palmitate was dissolved by methanol and filtered by 0.22滋m microfiltrationmembrane. The instrument condition: Using C18 reverse phase chromatography column,methanol and water (containing0.01% formic acid)were used as mobile phase,a flow rate of 0.8mL/min. The drift tube temperature was controlled at60℃ and the pressure of nitrogen at 25psi. Experimental results showed that the common logarithm for peak area andconcentration of ascorbyl palmitate showed good linear relationship (R2 =0.9972). The recovery ranged from 97.23% to102.05% and the relative standard deviation (RSD)was less than 5%. The method is accurate and rapid for quantifyingascorbyl palmitate content without derivatization.
